Test principle:This assay employs a two-site sandwich ELISA to quantitate PEPD in samples. An antibody specific for PEPD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPEPD present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for PEPD is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PEPD bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Xaa-Pro dipeptidase is a cytosolic dipeptidase that hydrolyzes dipeptides with proline or hydroxyproline at the carboxy terminus (but not Pro-Pro). It is important in collagen metabolism because of the high levels of aminoacids. encodes a member of the peptidase family. The protein forms a homodimer that hydrolyzes dipeptides or tripeptides with C-terminal proline or hydroxyproline residues. The enzyme serves an important role in the recycling of proline, and may be rate limiting for the production of collagen. Mutations in this gene resµLt in prolidase deficiency, which is characterized by the excretion of large amount of di- and tri-peptides containing proline. MµLtiple transcript variants encoding different isoforms have been found for this gene.
Stability:The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
